Why patients with familial hypercholesterolemia are at high cardiovascular risk? Beyond LDL-C levels

چکیده
Familial hypercholesterolemia (FH) is a common genetic cause of elevated low-density lipoprotein cholesterol (LDL-C) due to defective clearance circulating LDL particles. All FH patients are at high risk for premature cardiovascular disease (CVD) events their genetically determined lifelong exposure LDL-C levels. However, different rates CVD have been reported in patients, even among those with the same mutations and comparable Hence, additional modifiers, beyond LDL-C, may contribute increase population. In this review, we discuss overall burden Additionally, revise prognostic impact several traditional emerging predictors provide an overview role specific tools stratify order ensure them more personalized treatment approach.

LRP1 gene polymorphisms are associated with premature risk of cardiovascular disease in patients with familial hypercholesterolemia.
INTRODUCTION AND OBJECTIVES LRP1 gene overexpression in atherosclerotic plaque is associated with increased lipid uptake through the vascular wall. The aim of the study was to analyze whether LRP1 modulates the genetic risk of developing premature cardiovascular disease in familial hypercholesterolemia, using single nucleotide polymorphism association analysis. METHODS Ten polymorphisms of th...
متن کاملAntibody titers against oxidized LDL are not elevated in patients with familial hypercholesterolemia.
Antibodies against oxidized low density lipoproteins (Ox-LDLs) have been proposed to be independent predictors of atherosclerosis development. The main aims of the current study were to (1) compare antibody titers to Ox-LDL in patients with heterozygous familial hypercholesterolemia (n=51) with those in matched controls (n=45) and (2) analyze whether the antibody titers were related to the exte...
متن کاملPrevalence of high-risk cardiovascular patients with therapy-resistant hypercholesterolemia
INTRODUCTION Hypercholesterolemia is a causal risk factor for cardiovascular diseases, which is recommended to be treated at least in high-risk patients. Yet, currently there is a lack of epidemiological data on the number of high-risk patients in Germany who do not respond adequately to high-dose statin monotherapy or statin therapy in combination with other lipid-lowering agents. METHODS Of...
متن کاملPCSK9 R46L, lower LDL, and cardiovascular disease risk in familial hypercholesterolemia: a cross-sectional cohort study.
OBJECTIVE Proprotein convertase subtilisin/kexin type 9 (PCSK9) is a downregulator of the low density lipoprotein receptor. The aims of this cross-sectional cohort-study were to examine whether the PCSK9 R46L loss of function variant found in a cohort of familial hypercholesterolemia (FH) patients was associated with lower low density lipoprotein cholesterol, lower frequency of xanthomata, and ...
متن کاملGenetic Analysis of Iranian Patients with Familial Hypercholesterolemia
Background: Familial hypercholesterolemia (FH) is a frequent autosomal dominant disorder of lipoprotein metabolism. This disorder is generally caused by mutations in low-density lipoprotein receptor (LDLR), apolipoprotein B 100 (APOB), and proprotein convertase subtilisin/kexin type 9 (PCSK9) genes. In the present study, we aimed at identifying the common LDLR and APOB gene mutations in an Iran...
